If I have set some applications as iOS required and now I want to push them as VPP will the app be reinstalled?
I will for sure do the VPP assignment before I will remove the normal required configuration.

It depends how you do the assignments in Intune and the process you follow but form the iOS perspective no the app will not be re-installed as long as the config is done correctly.
